Proposed Analysis: | The proposed analysis will investigate interactions between polygenic risk for Alzheimer's Disease (AD) and clinical information in ADNI on smoking behavior or alcohol use to assess the degree to which genetic risk for AD is moderated by smoking behavior and alcohol consumption. In addition to polygenic scores for AD that are constructed using traditional methods, we will use tissue-specific scores that re-adjust polygenic score weights from genome-wide association studies for AD using functionally annotated data to improve understanding of biological pathways between smoking/alcohol consumption and AD. |
